Formulasi SNEDDS (Self-Nanoemulsifying Drug Delivery System) yang Mengandung Minyak Jagung (Oleum maydis)

Abstract. Corn oil contains 59% polyunsaturated fatty acids, 24% monounsaturated fatty acids, 13% saturated fatty acids, ubiquinone, and tocopherol. Corn oil is beneficial for lowering blood cholesterol levels. To maximize the absorption of corn oil, an effective delivery system is required, such as Self-Nanoemulsifying Drug Delivery System (SNEDDS). This study aims to formulate and evaluate corn oil SNEDDS using a D-optimal mixture design with variables including corn oil (X1), surfactant (X2), and cosurfactant (X3), and response variables consisting of emulsification time (Y1) and percent transmittance (Y2). The optimum formula was determined based on a short emulsification time and high percent transmittance. The optimal formula was found in formulation 4, composed of 10% corn oil, 50% Tween 80, and 40% PEG 400. Evaluation results showed a globule size of 7.233 ± 0.321 nm with a polydispersity index of 0.171 ± 0.132, a zeta potential of -0.1 ± 1.699 mV, emulsification time of 18.076 ± 1.091 seconds, and percent transmittance of 98.816 ± 1.800. No phase separation occurred in the robustness test, and the formula remained stable during both heating-cooling and centrifugation stability tests.
Abstrak. Minyak jagung mengandung 59% asam lemak tak jenuh ganda, 24% asam lemak tak jenuh tunggal, 13% asam lemak jenuh, ubikuinon, dan tokoferol. Minyak jagung bermanfaat untuk menurunkan kadar kolesterol dalam darah, untuk memaksimalkan penyerapan minyak jagung maka diperlukannya sistem penghantaran yang efektif salah satunya seperti SNEDDS. Penelitian ini bertujuan untuk merumuskan dan mengevaluasi SNEDDS minyak jagung menggunakan D-optimal mixture design dengan variable berupa minyak jagung (X1), surfaktan (X2), dan kosurfaktan (X3), serta variable respon berupa waktu emulsifikasi (Y1) dan persen transmitan (Y2). Formula optimum ditentukan dari hasil waktu emulsifikasi rendah dan persen transmitan tinggi. Formula optimum terdapat pada formula 4 dengan komposisi minyak jagung 10%, Tween 80 50%, dan PEG 400 40%, dengan hasil evaluasi diantaranya memiliki ukuran globul 7.233 ± 0.321 nm dengan nilai indeks polidispersitas sebesar 0.171 ± 0.132, memiliki nilai zeta potensial sebesar -0.1 ± 1.699 mV, waktu emulsifikasi 18.076 ± 1.091 detik, nilai persen transmitan sebesar 98,816 ± 1,800, tidak terjadi pemisahan fasa berdasarkan uji robustness, dan stabil pada uji stabilitas baik heating cooling maupun sentrifugasi.
