The central and peripheral analgesic effect of Vietnamese traditional herbal formula Thai Binh HV decoction on experimental animal models

Background: The extract of Thai Binh HV (TBHV), a traditional Vietnamese herbal formula, has been found to be safe and anti-arthritis effects, but the mechanism of analgesic action of this formula has not been clarified as yet. Objectives: This study aimed to investigate the central and peripheral analgesic effects of TBHV. Materials and Methods: The study was conducted on Swiss mice and Wistar white rats to evaluate the analgesic effect of TBHV with three models corresponding to different pain agents (hot plate, acetic acid-induced writhing, and carrageenaninduced paw edema). Control drugs in the study included oral codeine phosphate dose 20 mg/kg, aspegic dose 180 mg/kg, and diclofenac sodium 15 g/kg. The research product TBHV was used orally at doses of 20.4 g/kg and 40.8 g/kg for mice and at doses of 11.9 g/kg and 23.8 g/kg for rats. Results: The remedy TBHV demonstrated effective analgesic effects in experimental models. In the hot plate model, both doses (20.4 g/kg/day and 40.8 g/kg/day) displayed significant analgesic effects. When tested using the Koster method, the remedy exhibited notable analgesic effects by reducing writhing symptoms compared to the control group, equivalent to the analgesic impact of aspegic at a dose of 180 mg/kg/day. In the carrageenan-induced paw edema test, oral administration of TBHV at doses of 11.9 g/kg and 23.8 g/kg in rats produced an analgesic effect by increasing the pain threshold. Conclusion: TBHV has exhibited evident analgesic effects through both central and peripheral mechanisms.
