Interaction of Phenytoin and Folate in the Rat

Summary: The interactions between folate and phenytoin were studied in the rat using a model in which constant, nontoxic, and continuously protective levels of phenytoin were maintained. After 10 days of treatment with phenytoin, liver folate concentration was decreased while brain, plasma, and adrenal folate concentrations remained unaffected. Oral folate supplementation (20 mg/kg) increased folate concentrations in all tissues examined in phenytoin‐treated animals, but had no effect on phenytoin levels. Folate supplementation did, however, increase the recovery time after maximal electroshock seizures in phenytoin‐treated rats, but did not influence phenytoin's ability to protect against tonic hindlimb extension. Oral folate supplementation in animals not treated with phenytoin also significantly increased folate concentrations in all tissues examined except brain.RÉSUMÉLes interactions entre folate et phénytoine ont étéétudiées sur le rat grâce à un modele chez qui des niveaux efficaces, constants et non toxiques de phénytoine étaient maintenus en permanence. Aprés 10 jours de traitement par la phénytoine la concentration en folate diminuait dans le foie alors qu'elle restait in‐changée dans le cerveau, le plasma et les surrenales. Un supplément oral de folate (20 mg/kg) en augmentait les concentrations dans tous les tissus examinés chez les animaux traités par phénytoine, mais n'avait pas D'effet sur les niveaux de phénytoine. La supplémentation en folate, cependant, augmentait le temps de récupération aprés des crises MES chez les rats traités par la phénytoine, mais n'avait pas D'influence sur le pouvoir de protection de la phénytoine contre L'exten‐sion tonique des membres inféieurs. Sin embargo el suplemento de folato aumentó el tiempo de recuperación de los ataques MES en las ratas tratadas con fenitoina pero no influenceó la capacidad de la fenitoina de protección contra la extensión tónica de las patas posteriores. Verwendung fand ein Modell, bei dem konstante, nicht toxische, ständig protektive Spiegel des Phenytoins aufrecht erhalten wurden. Nach 10‐tägiger Behandlung mit Phenytoin war die Folat‐Konzentration in der Leber vermindert, während sie im Gehirn, im Plasma und in den Neben‐nieren unbeeinflusst blieb. Orale Folat‐Gabe (20 mg pro KG/kg) erhöhte die Folatkonzentrationen in alien untersuchten Geweben bei den Phenytoin‐behandelten Tieren; sie hatte keine Auswirkungen auf den Phe‐nytoin‐Spiegel. Die Folat‐Gabe erhöhte zwar die Er‐holungszeit nach MES‐Krämpfen bei Phenytoin‐behandelten Ratten, beeinflusste jedoch nicht die Schutzfunktion des Phenytoins gegeniiber tonischer Extension der Hinterpfote. Die orale Folat‐Verfütte‐rung an Tiere, die nicht mit Phenytoin behandelt wurden, erhöhte die Folatkonzentrationen in alien untersuchten Geweben mit Ausnahme des Gehirns ebenso signifikant.
