Applications of Galβ1-3GalNAc Lectins in Cancer Diagnostics and Therapy

Aberrant glycosylation is a hallmark of cancer, prominently featuring the exposure of truncated O-glycans such as the Thomsen-Friedenreich (T) antigen (Galβ1-3GalNAc). Lectins that recognize this structure, including Peanut Agglutinin (PNA) and Amaranthus caudatus lectin (ACL), have emerged as powerful tools in cancer diagnostics and therapeutic targeting. This article reviews the diagnostic applications of T-antigen-binding lectins in histopathology, serum biomarker discovery, and prognostication, alongside emerging therapeutic strategies targeting galectin-mediated interactions, particularly galectin-3. These approaches highlight the translational potential of glycan-lectin biology in precision oncology.
