Organizmalar, DNA hasarının olumsuz sonuçlarını azaltmak için birlikte işlev gören sistemlerle (DNA onarımı/tamiri, hasar toleransı, hücre döngüsü kontrol noktaları ve hücre ölüm yolları) donatılmıştır. Hücre DNA tamir stratejisi olarak dört farklı yol izler; (a) Direkt tamir sistemleri, (b) Hasarlı bölümün çıkartılmasını takiben yeni sentez tamir (Kesip-Çıkarma Tamir Sistemleri), (c) Çift zincir kırıklarının tamiri (Homolog rekombinasyon ve Nonhomolog uç birleştirme), (d) Translesion DNA Sentezi (DNA Hasar Toleransı). DNA tamir mekanizmasındaki yetersizlik veya bozukluklar, bazı kanser ve hastalıklara yol açabildiği gibi otosomal ressesif kalıtım gösteren DNA tamir bozuklukları ile ilişkili sendromlar vardır; Xeroderma Pigmentosum, Ataxia Telangiectasia, Fanconi Anemia.
