Cuprizone

Abstract Cuprizone is a disubstituted hydrazine with a MP of 214°-2160C. It is a copper chelator and has been used in a quan titative spectrophotometric procedure for copper analysis (17). In a study of an animal model of swayback (a copper deficiency disease of sheep and calves), cuprizone caused hydrocephalus, spongiform change, demyelination, and an Alzheimer’s type II astrocytosis in the CNS of weanling mice (6). The spongiform change was due to intramyelinic edema, and giant mitochondria and proliferated smooth en doplasmic reticulum appeared in hepatocytes (19). Remye lination occurs quickly after cuprizone is withdrawn from the diet, making cuprizone a useful neurotoxicant for pro ducing experimental models of demyelination and remye lination in the CNS (2-4,14). There are no reports of cu prizone toxicity in humans. The absorption, distribution, metabolism, and excretion of cuprizone are unknown. The CNS, PNS and liver are target organs in experimental animal models.
