Characterization of SWS1-SWSAP1 : the human Shu complex

Caractérisation de SWS1-SWSAP1 : le complexe Shu humain La préservation de l’intégrité de l’ADN est essentielle pour la stabilité du génome. Les cassures du double-brin d’ADN sont un des types de dommages à l’ADN les plus sévères. Elles peuvent être réparées par un mécanisme de réparation utilisant une séquence d’ADN homologue (HDR) via des échanges catalysés par RAD51, de la famille de recombinase Rec-A. RAD51 agit comme un filament nucléoprotéique catalysant la reconnaissance d’homologie et l’échange de brin, contrôlés par des modulateurs. Récemment, SWSAP1, qui interagit avec SWS1 et SPIDR pour former le «complexe SHU» ont été identifiés comme modulateurs de RAD51. Cependant, il reste à déterminer comment le complexe SHU contrôle l’activité de RAD51.Dans les cellules somatiques, le complexe SHU est requis pour l’assemblage stable de RAD51 aux sites de dommages de l’ADN, mais il est étonnamment non-essentiel pour le HDR intra-chromosomal. Cependant, SWS1-SWSAP1-SPIDR apparait essentiel pour le HDR inter-homologues, une voie de réparation active dans les cellules germinales mais normalement réprimée dans les cellules somatiques. Afin de mieux comprendre le mécanisme d’action du complexe SHU dans le contrôle de RAD51, mon projet de thèse a pour but de caractériser les activités biochimiques de SWSAP1. Premièrement, j’ai analysé les interactions de SWSAP1 avec SWS1, SPIDR, RAD51 et BLM à l’échelle protéique. Ensuite, j’ai étudié l’interaction de SWS1-SWSAP1 avec l’ADN. Pour terminer, j’ai examiné l’impact de SWS1-SWSAP1 sur la dynamique et les activités du filament nucléoprotéique de RAD51. D’après les résultats de mes travaux de thèse, j’expose de potentiels modèles pour la fonction du complexe Shu dans le contrôle de RAD51.
