Uji Aktivitas Antibakteri Ekstrak Eksokarp Buah Semangka Merah (Citrullus lanatus (Thunb.) Matsum & Nakai) terhadap Bakteri Propionibacterium acnes

Abstract. Acne vulgaris is a chronic inflammation of the pilosebaceous follicles caused by various factors, including genetics, hormones, diet, and environmental influences. One of the primary bacteria responsible for acne is Propionibacterium acnes. The use of conventional antibiotics to treat acne often leads to bacterial resistance. Therefore, this study aims to investigate the potential of watermelon (Citrullus lanatus) exocarp extract as a natural antibacterial alternative against Propionibacterium acnes. This research employs the maceration extraction method using 96% ethanol as a solvent to obtain active compounds from the watermelon exocarp. The antibacterial activity test was conducted using the agar well diffusion method against Propionibacterium acnes. The results showed that the watermelon exocarp extract exhibited inhibitory effects on Propionibacterium acnes, with moderate inhibition zones at concentrations of 20%, 21%, 25%, and 30%, measuring 8.1 mm, 8.5 mm, 9 mm, and 10.6 mm, respectively. Phytochemical analysis identified active compounds such as saponins, alkaloids, polyphenols, tannins, and flavonoids, which contribute to antibacterial activity. In conclusion, this study suggests that watermelon exocarp extract has potential as a natural antibacterial agent against Propionibacterium acnes and may be developed as an alternative natural acne treatment.
Abstrak. Acne vulgaris merupakan peradangan kronis pada folikel polisebasea yang disebabkan oleh berbagai faktor, seperti genetik, hormon, diet, dan lingkungan. Salah satu bakteri utama penyebab jerawat adalah Propionibacterium acnes. Penggunaan antibiotik konvensional untuk mengatasi jerawat seringkali menimbulkan resistensi bakteri. Oleh karena itu, penelitian ini bertujuan untuk mengkaji potensi ekstrak eksokarp buah semangka (Citrullus lanatus) sebagai alternatif antibakteri alami terhadap Propionibacterium acnes. Penelitian ini menggunakan metode ekstraksi maserasi dengan pelarut etanol 96% untuk memperoleh senyawa aktif dari eksokarp buah semangka. Uji aktivitas antibakteri dilakukan dengan metode difusi agar cara sumuran terhadap Propionibacterium acnes. Hasil penelitian menunjukkan bahwa ekstrak eksokarp buah semangka memiliki daya hambat terhadap Propionibacterium acnes dengan zona hambat berkategori sedang pada konsentrasi 20%; 21%; 25% dan 30%, dengan zona hambat 8,1mm; 8,5mm; 9mm dan 10,6mm. Analisis fitokimia mengidentifikasi senyawa aktif seperti saponin, alkaloid, polifenol, tanin, dan flavonoid, yang berperan dalam aktivitas antibakteri. Kesimpulan penelitian ini menunjukkan bahwa ekstrak eksokarp buah semangka memiliki potensi sebagai antibakteri alami terhadap Propionibacterium acnes yang dapat dikembangkan sebagai alternatif obat antijerawat berbasis bahan alami.
