Polimer Semi-Sintetik Berbasis Karbopol, HPMC, dan Na-CMC untuk Patch Mukoadhesif

Abstract. Bukal mucoadhesive patches are an important innovation in drug delivery as they enable rapid drug absorption and bypass first-pass metabolism, thereby improving therapeutic efficacy and patient compliance. The success of a mucoadhesive patch is strongly influenced by the choice of polymer matrix, which plays a crucial role in film formation, mucoadhesion, and controlling the drug release rate. This narrative literature review examines scientific articles published between 2015 and 2025 that were retrieved from PubMed, Scopus, ScienceDirect, and SpringerLink. The review evaluates the characteristics, advantages, and optimal combinations of the semi-synthetic polymers Carbopol, HPMC, and Na-CMC in bukal mucoadhesive patch formulations. Analysis of the literature indicates that each polymer has a specific role: Carbopol and Na-CMC provide high mucoadhesive strength, prolonging the patch’s residence time on the mucosal surface, whereas HPMC forms a stable film matrix and modulates drug release in a controlled manner. The combination of all three polymers synergistically enhances patch performance, balancing prolonged adhesion with a controlled drug release profile. In conclusion, an optimized formulation combining Carbopol, HPMC, and Na-CMC yields a bukal mucoadhesive patch that is effective and stable, with controlled drug release and high mucoadhesive strength.
Abstrak. Patch mukoadhesif bukal merupakan inovasi penting dalam sistem penghantaran obat karena memungkinkan penyerapan obat yang cepat dan menghindari metabolisme lintas pertama, sehingga meningkatkan efektivitas terapi dan memenuhi pasien. Keberhasilan patch mukoadhesif sangat dipengaruhi oleh pemilihan polimer matriks yang berperan dalam membentuk film, melekat pada mukosa, dan mengontrol laju pelepasan obat. Studi literatur naratif ini mengkaji artikel ilmiah terbitan 2015–2025 yang ditelusuri melalui database PubMed, Scopus, ScienceDirect, dan SpringerLink untuk menghasilkan karakteristik, keunggulan, dan kombinasi optimal polimer semi-sintetik Karbopol, HPMC, dan Na-CMC dalam formulasi patch mukoadhesif. Hasil analisis menunjukkan bahwa masing-masing polimer memiliki peran spesifik: Karbopol dan Na-CMC memberikan kekuatan mukoadhesi tinggi yang memperpanjang waktu kontak patch dengan mukosa, sedangkan HPMC membentuk matriks film yang stabil dan mengatur pelepasan obat secara terkendali. Kombinasi ketiga polimer secara sinergis meningkatkan kinerja patch , menyeimbangkan daya lekat jangka panjang dengan profil pelepasan obat yang terkontrol. Kesimpulannya, formulasi patch mukoadhesif bukal dengan kombinasi Karbopol, HPMC, dan Na-CMC yang optimal menghasilkan sediaan yang efektif dan stabil serta mampu melepaskan obat secara terkendali dengan daya lekat mukosa yang tinggi.
