Formulation of keratin/HPMC matrix tablets for sustained delivery of diclofenac potassium for the management of inflammation

Purpose: To investigate the excipient properties of keratin/HPMC in sustained-release diclofenac matrix tablets.Method: Tablets were prepared by wet granulation using different keratin/HPMC ratios. The granules were evaluated for pre-compression parameters such as flow properties, Hausner’s ratio, Carr’s index, while the matrix tablets were evaluated for post-compression parameters such as hardness, disintegration time, in-vitro drug release and release kinetics.Results: There was no significant difference (p > 0.05) in flow properties and compression characteristics in formulations containing either keratin alone or in combination with HPMC. Furthermore, formulations containing keratin/HPMC in equal proportions exhibited better sustained release profiles (p < 0.05). The FTIR spectra revealed no change in the functional groups of diclofenac potassium.Conclusion: Combination of keratin/HPMC improves the sustained release profile of diclofenac matrix tablets compared to either ingredient alone.
