Objectives
The observe the effects of curcumine on the myocardial cells of neonatal rats injured by hypoxiareoxygenation. Whether the protective effect related with heat shock protein 70 (HSP70) expression.
Methods
Primary culture myocardial cells from neonatal Sprague Dawley (SD) rats rat. Through the MTT method measuring IC50 sure curcumin concentration. Established an anoxia/reoxygenation model and measured the activity of lactate dehydrogenase (LDH), the content of malonaldehyde (MDA). Analysis the cell apoptosis and necrosis and used immunohistochemistry detection HSP70 expression.
Results
LDH activity and MDA content in different doses curcumin group changed different with time. Myocardial cells apoptosis and necrosis and HSP70 expression in connection with curcumin doses and function about time.
Conclusions
LDH activity and MDA content in different doses curcumin group changed different with time. Myocardial cells apoptosis and necrosis and HSP70 expression in connection with curcumin doses and function about time. Curcumine could protect cultured neonatal cardiomyocytes from hypoxia/reoxygenation injured. And the protect effect was dose-dependently. The protective effects of curcumin on hypoxia/reoxygenation myocardial cells may be related with the increasing of HSP70 expression.
