COMPARATIVO ENTRE WESTERN BLOTTING E QUIMIOLUMINESCÊNCIA (CLIA) NA DETECÇÃO DO AUTOANTICORPO ANTI-LEDGF75/DFS70 EM SOROS DE PACIENTES

Objetivo: Este trabalho buscou comparar a sensibilidade e a especificidade de duas diferentes metodologias, Western Blotting por revelação colorimétrica e Quimioluminescência (CLIA), para a detecção do autoanticorpo anti-LEDGF75 (lens epithelium-derived growth fator p75 ou DFS70) em soro, associando também ao resultado de FAN (fator antinúcleo por imunofluorescência indireta em Hep-2). Método: Foram processadas as amostras de 129 pacientes, no período de um ano, em ambas as metodologias, assim como na pesquisa de FAN por imunofluorescência indireta em HEp-2. Primeiramente, comparou-se CLIA × W.BLOT e, posteriormente, W.BLOT × FAN e CLIA × FAN. Entre os 58 casos negativos no Western Blot, houve concordância de 89,6% com o resultado por CLIA; e entre os 71 casos positivos no Western Blot, houve concordância de 97,1% com o resultado por CLIA. Posteriormente, associou-se ao padrão de FAN, especialmente com o padrão nuclear pontilhado fino denso (AC-2), esperado para os casos reagentes de anti-LEDGF75/DFS70. Conclusão: Conclui-se que é viável deixar de detectar o autoanticorpo AALEDGF75/DFS70 pelo método Western Blotting, que é mais vagaroso e subjetivo, e substituí-lo pelo método de quimioluminescência (CLIA), o qual apresentou resultados confiáveis, concordantes e quantificáveis, sendo seu processamento mais fácil e seu sistema eficaz de rastreabilidade e interfaceamento dos resultados.
