Phytochemical study of in vitro cultured Sweet flag (Acorus calamus Linn.,)

Acorus calamus, registered in the Mongolian Red book as an endangered, is a perennial herb that synthesizes about 140 bio-active compounds including phenylpropanoids, flavonoids, mono, and sesquiterpenes. The most common ingredient in the prescriptions for Mongolian traditional medicine, sweet flag (Acorus calamus) has been used extensively in traditional oriental medicine and is still used in medicine, food and cosmetic industries in many countries today. Due to shrinking of natural habitat and distribution area of the species, it is required to generate the raw materials for the production of medicines and other final products as a result of propagating them through advanced technology in laboratory conditions. To confirm whether we could substitute application of the natural population through the in vitro propagated plantlets, it is also necessary to compare their content of predominant compounds with natural grown plants. In this study, A.calamus L. were propagated by tissue culture methods, adapted in ex vitro condition and transferred to the open field (in vivo), and the secondary metabolites in the plantlets at every step of culture (in vitro, ex vitro, in vivo) were identified through the gas chromatography-mass spectrometry (GC-MS). The samples from the leaves, rhizomes, and roots were extracted in n-hexane and performed quantitative determination of phytochemical constituents. Totally 55 compounds were found in the samples such as methylisoeugenol (0.05-9.18%), shyobunone (0.03-1%), acoradiene (0.05-2.86%), α-pinene (0.09-0.51%), β-selinene (0.26-6.64%). In addition, 33 mono and sesquiterpene compounds including α-pinene, camphene, linalool, geranyl acetate, β-farnesene, curcumin, and nerolidol were detected in the essential oils extracted from ex vitro rhizome through the phytochemical quality analysis. Дорнын уламжлалт анагаах ухаанд нэн эртнээс хэрэглэж ирсэн эмийн болон экологийн өндөр ач холбогдолтой Эгэл годил өвсийг дэлхийн олон оронд эм, хүнс, гоо сайхны үйлдвэрлэлд ашигладаг бөгөөд Монгол орны уламжлалт эм тангийн жоронд орох давтамжаар тэргүүлдэг. Эгэл годил өвсний тархац нөөц багасаж, байгаль дээр сэргэн ургах боломж буурсан өнөө үед орчин үеийн дэвшилтэт технологи ашиглан лабораторийн нөхцөлд олшруулан тарималжуулж, тэдгээрийн ашигт бодисын агууламжийг байгалийн ургамалтай харьцуулан тодорхойлсноор эм болон бусад эцсийн бүтээгдэхүүний үйлдвэрлэлийн түүхий эдийн нөөц бүрдүүлэх боломжтой юм. Энэхүү судалгаагаар Эгэл годил өвсийг эдийн өсгөврийн аргаар олшруулан, хөрсөнд дасгаж, улмаар бичил ургамалд агуулагдах хоёрдогч метаболитын агууламжийг хийн хроматограф-масс спектрометрийн аргаар тодорхойлсон. Ургамал тус бүрийн навч, үндэс, үндэслэг ишний ялгаатай дээжүүдийг гександ хандлан фитохимийн тоон тодорхойлолт хийхэд голлох нэгдлүүд болох метилизоэвгенол (0.05-9.18%), шиобунон (0.03- 1%), акорадиен (0.05-2.86%), α-пинен (0.09-0.51%), β-селинен (0.26-6.64%) болон неролидол, аромадендрен зэрэг нийт 55 нэгдэл илэрсэн.
