Celastrus Paniculatus Willd (Malkangni): A Comprehensive Review on Phytochemistry, Biological Activities, Pharmacological Mechanisms, and Clinical Advancements

Celastrus paniculatus Willd. (Family: Celastraceae), commonly known as Malkangni, Jyotishmati, or the Intellect Tree, is a perennial woody climbing shrub widely distributed across the Indian subcontinent and Southeast Asia. This plant holds a pre-eminent position in Ayurvedic, Unani, and Siddha traditional medicine systems, where it has been prescribed for centuries as a "Medhya Rasayana" (nervine tonic and cognitive enhancer). The seeds and seed oil of C. paniculatus are the primary therapeutic parts, containing a rich array of bioactive phytochemicals including sesquiterpenoid polyalcohols and esters (malkanguniol, malkangunin, celapanin, celapnin), alkaloids (celastrine, paniculatine), phenolic triterpenoids (celastrol, paniculatadiol), polyunsaturated fatty acids (linoleic, linolenic, oleic, palmitic acids), agarofuran derivatives, and flavonoids. Extensive preclinical investigations have validated the plant's multiple pharmacological activities including neuroprotective, cognition enhancing, antioxidant, anti-inflammatory, antimicrobial, antianxiety, antidepressant, anticonvulsant, antifertility, hypolipidemic, anticancer, wound healing, antimalaria, and anti-arthritic effects. The seed oil modulates key neurotransmitter systems (acetylcholine, dopamine, and serotonin), suppresses NF-κB-mediated neuroinflammation, reduces oxidative stress via antioxidant enzyme augmentation, and exhibits neuroprotection in diverse animal models. However, clinical trial data in humans remains limited. This review comprehensively consolidates the current knowledge on botanical characteristics, ethnopharmacology, phytochemistry, biological activities, pharmacological mechanisms, and clinical status of C. paniculatus, highlighting future research imperatives and conservation challenges.
