Reaction of MM Red Cells with Vicia Graminea Extract, Anti‐N: Qualitative Differences Between Capillary and Venous Blood Samples

SummarySaline extracts of a batch of Vicia graminea seeds were found to react strongly at 21°C with saline suspensions of red cells from fresh venous samples of MM genotype. Capillary samples from the same donors were not agglutinated. The reaction was consistent and reproducible and was shown by clotted and anticoagulated venous samples. It persisted after the venous red cell samples had been stored for 12 months in 40% glycerol at ‐25°C. The reaction appeared to be independent of the presence of serum in the test system and of the pH of the sample or suspending medium. Of venous samples from 23 MM donors tested, 15 reacted with Vicia extract. There was a high proportion of MSMS and MSMs genotype within this group and of MsMs genotype among the 8 non‐reactors. Capillary and venous red cells from two of the reacting donors were unagglutinated by human or rabbit anti‐N and gave a normal “double‐dose” score with horse anti‐M.All extracts of this batch of seeds made this qualitative distinction. It has not been encountered in the use of anti‐N from human or animal sources nor extracts of other batches of Vicia graminea seeds.RésuméDes extraits en solution physiologique d'un lot de semences de Vicia graminea se sont avérés avoir la propriété de réagir fortement à la température de 21° avec une suspension érythrocytaire en solution physiologique provenant de sang veineux frais de génotype MM. Les échantillons de sang capillaire provenants des mêmes donneurs ne sont pas agglutinés. La réaction est constante et reproductible et se manifeste aussi bien avec le sang veineux coagulé qu'avec le sang veineux non coagulé. Elle persiste après un stockage de 12 mois à moins 25° C lorsque les échantillons d'érythrocytes veineux sont conservés en solution glycérinée à 40%. Lors des tests, la réaction semble être indépendante de la présence de sérum et du pH de l'échantillon ou du milieu de suspension. Parmi 23 échantillons veineux de donneurs MM, 15 réagissent avec l'extrait de Vicia. Parmi ceux qui réagissent, on observe une haute proportion de personnes de génotypes MSMS et MSMs; par contre, parmi les 8 personnes qui ne réagissent pas, on observe une grande proportion de personnes de génotype MsMs. Les érythrocytes de sang veineux et capillaire provenant de 2 donneurs présentant la réaction ne sont pas agglutinés par les anti‐N d'origine humaine ou de lapin et donnent une réaction normale de double dose avec l'anti‐M de cheval.Tous les extraits de ce lot de semences font cette distinction qualitative. Die Reaktionen waren sowohl bei Verwendung geron‐nener wie auch mit Antikoagulanslösungen versetzter Venenblutproben eindeutig und reproduzierbar. Die Reaktion war selbst bei Venenblutproben, die während 12 Monaten in 40% Glyzerin bei ‐25° C eingefroren waren, noch eindeutig nach‐weisbar. Für die Reaktion war die Anwesenheit von Serum unnötig; ebensowenig wurde sie durch das pH der Blutprobe oder der Aufschwemmungsflüssigkeit beeinflußt. 15 von 23 Venenblutproben von MM‐Spendern zeigten im Ansatz mit dem Vicia‐Extrakt eine positive Reaktion. In dieser Gruppe fand sich ein übergroßer Anteil der Genotypen MSMS und MSMs. Bei den 8 Proben mit negativer Reaktion wurde hingegen eine Häufung des Genotyps MsMs beobachtet. Die Kapillar‐ und Venenblutproben von zwei positiven Spendern wurden durch menschliche und Kaninchen‐Anti‐N‐Seren nicht agglutiniert; mit einem Pferde‐Anti‐M zeigten sie einen normalen Doppeldosen‐Score.Sämtliche Extrakte des betreffenden Samenmusters zeigten die erwähnte Eigenheit. Sie wurde bei andern Mustern von Vicia graminea‐Samen sowie bei Anti‐N‐Seren menschlicher oder tierischer Herkunft nie beobachtet.
