Duodenal Bulb Motor Activity in Sheep

SummaryRecordings of the parietal contractile force and electrical activity of the ovine gastroduodenal junction indicated the ability of the cranial part of the duodenum to function as a “reservoir” cyclically evacuated in front of serial contractions. These serial contractions reached the proximal duodenum as phases III of the migrating myoelectric complex. Their daily number was correlated with the amount of digesta, e. g. lowered during a 48‐h fast period and increased at refeeding, and whith age (as the younger the animal the higher the daily number of complexes).Premature serial contractions from the duodenal bulb and phases III were evoked by several treatments, including 5‐HT and its precursor, 5‐HTP. However, they only maintained cycling at a high frequency after the administration of the 5‐HT antagonist, methysergide.It is concluded that the ovine duodenal bulb functions as an independent unit which involves myenteric serotoninergic neurones in the initiation of the cyclical motor activity of the intestine. The increased number of cycles caused by methysergide suggests a permissive effect by blocking an age‐dependent 5‐HT inhibitory mechanism.ZusammenfassungDie Funktion des Bulbus duodenalis beim SchafDie Aufzeichnungen der Kontraktionskräfte und der elektrischen Aktivität des gastroduodenalen Übergangs beim Schaf zeigten, daß der proximale Teil des Duodenums, der Bulbus duodeni, eine Reservoirfunktion hat. Dieser Teil wird zyklisch vor einer Serie von Kontraktionen entleert, die der Phase III des myoelektrischen Komplexes entsprechen. Die Anzahl dieser Phasen III steht in einem festen Verhältnis zum Volumen der Mahlzeiten: Sie ist nach einer Hunger‐Periode von 48 Stunden vermindert und steigt bei Realimentation wieder an. Sie ist außerdem bei jungen Tieren höher als bei erwachsenen Tieren.Transitorische Kontraktionserien am Bulbus und korrespondierende Phasen III konnten durch zahlreiche Substanzen, darunter auf 5‐HTP und dessen Metabolit 5‐HT, provoziert werden. Unter den Antagonisten des 5‐HT bewirkte das Methysergid eine starke Erhöhung der Frequenz der Phasen III während mehrerer Stunden bei erwachsenen Tieren.Der Bulbus duodeni scheint also die Funktion einer unabhängigen Einheit mit den myenterischen, serotoninergen Neuronen am Ursprung der zyklischen motorischen Aktivität des Interstitium zu haben.
