Effects of Vigabatrin (γ‐vinyl GAB A) on Neuro transmission‐Related Amino Acids and on GAB A nd Benzodiazepine Receptor Binding in Rats

Summary: The effect of 12‐day intraperitoneal i.p. administration of vigabatrin (GVG, γ‐vinyl GAB A) to rats on the neurotransmission‐related amino acids in various brain regions (cortex, hippocampus, cerebellum, and spinal cord), cisternal fluid (CSF) and blood was studied. Results showed that GVG administration increased the levels of GABA in cortical and subcortical regions of the brain and CSF without affecting GABA and benzodiazepine receptors in the cortex. In addition, a dose‐dependent decrease was noted in the concentration of gluta‐mate in the hippocampus and in the concentrations of aspartate and glutamine in the cortex, hippocampus, and cerebellum. The changes in the levels of amino acids in the brain, except for that of GABA, were not reflected in the CSF, however, and the levels of amino acids in discrete brain regions did not show any correlation with those in the serum or in the CSF. The results suggest that GVG administration might suppress development and spread of seizures not only by elevating the level of the inhibitory amino acid GABA, but also by decreasing the levels of excitatory amino acids in the brain.RÉSUMÉLes effets ďune administration intrapéritonéale de Vigabatrin (γ‐Vinyl GABA, GVG) pendant douze jours chez le rat sur les amino‐acides neurotransmetteurs au niveau de différentes régions cérébrales (cortex, hippocampe, cervelet, moëlle épiniére) au niveau du LCR et au niveau du sang ont étéétudiés dans ce travail. Les résultats montrent que ľadministration de GVG augmente les taux de GABA au niveau des régions corticales et sous‐corticales du cerveau et au niveau du LCR, sans avoir ďeffets sur les récepteurs GABA et benzodiazépines au niveau du cortex. De plus, une diminution dose‐dépendante a été constaée pour les concentrations de glutamate dans ľhippo‐campe, et pour les concentrations ďaspartate et de glutamine au niveau du cortex, de ľhippocampe et du cervelet. Cependant les modifications des taux ďamino‐acides au niveau du cerveau (avec ľexception de celui de GABA) ne se traduisaient pas au niveau du LCR; les taux ďamino‐acides au niveau des différentes régions cérébrates n'ont pas été corrélés avec les taux sériques ou les taux du LCR. Ces résultats suggérent que ľad‐ministration de GVG pourrait supprimer le développement et ľextension de crises, non seulement en élevant les taux de GABA, acide aminé inhibiteur, mais également en diminuant les taux des acides aminés excitateurs au niveau du cerveau.RESUMENSe ha estudiado el efecto de la administración intraperitoneal durante 12 días, en ratas, de vigabatrin (GVG, gamma‐vinyl GABA) sobre la neurotransmisión por aminoácidos en varias regiones cerebrales (corteza, hipocampo, cerebelo y médula es‐pinal), en h'quido céfalo‐raquídeo (CSF) y en sangre. Los resulta‐dos muestran que la administración de GVG incrementan los niveles de GABA en las regiones corticales y subcorticales del cerebro y el CSF sin afectar el GABA ni los receptores de ben‐zodiazepina en la corteza. Además se encontró una reductión dosis‐dependiente de la concentrateón de glutamato en el hipocampo y de las concentraciones de aspartato y glutamina en la corteza, hipocampo y cerebelo. Sin embargo, los cambios en los niveles de aminoácides en el cerebro, con exceptión de los de GABA, no se reflejaron en el CSF ni los níveles de aminoácidos detectados en discretas regiones cerebrales mostraron correlateón alguna con los del suero o los del CSF. Estos resultados sugieren que la administración de GBG pueden suprimir el de‐sarrollo y la propagateón de los ataques no sólo elevando los niveles del aminoácido inhibitorio, GABA, sino también re‐duciendo los niveles de los aminoácidos excitatorios en el cerebro.
